The itinerary is as follows:

Take the JR Kuroshio train from Shin-Osaka Station to Wakayama Station. The journey will last about 1 hour.

From there, take the Nankai Koyasan Cable Car for a short 5-minute ride. Get off at Koyasan station and you will have arrived. From there you will have to walk about 2.3 km until you reach your destination. It will take approximately 40 minutes to reach the Buddhist area from Koya Station.
